Abstract

Biochemical and molecular techniques, isozyme pattern and random amplified polymorphic DNA (RAPD) analysis were applied in sorghum cultivars analysis to compare suitability for quantifying genetic diversity. Considering the limits of the use of morphological markers, the reliable estimate of DNA fingerprinting of the sorghum cultivars was assessed using isozyme banding pattern and RAPD analysis using random operon primers. Random amplified polymorphic DNA (RAPD) technique was used for genetic analysis of nine sorghum cultivars with different eight random operon primers. All eight operon primers were polymorphic and 67 amplified bands were obtained, out of which 44 were polymorphic, accounting 65.67% polymorphism. OPC 19 was most informative primer with 10 polymorphic bands, which can be used for further study of sorghum germplasm. Thus the results obtained on the polymorphism with different operon primers in relation to cultivars and the known pedigree information indicate the usefulness of the RAPD marker in determining genetic distance in sorghum. Peroxidase banding pattern was very prominent at Rf values 0.36, 0.50, 0.76 and 0.80. Banding patterns of peroxidase isozymes for M 35-1 and Selection - 3 was very dark than other genotypes. This type of information can be helpful in basic identification of molecular differences in the various types of sorghum genotypes.
