Abstract

In an attempt to standardize the X-ray contrast method, various cereal and millet seeds were subjected to different soaking and 10 or 20% BaCl2 treatments. A 2-h treatment of the presoaked seeds of wheat, jowar (Sorghum bicolor) and pearl millet (Pennisetum americanum) with 20% BaCl2 and a 4-h treatment of maize seed, also with 20% BaCl2, gave the best contrast between impregnated (dead) and unimpregnated (viable) seeds. Other smaller millets (Eleusine coracana and Paspalum scrobiculatum) required treatment for 4 h after presoaking for 16 h, and in oats barley and rice a clear impregnation was not obtained. Alternative contrast agents were evaluated and it is suggested that the contrast is emphasized by slight over-developing of X-ray plates or treatment with lactophenol after BaCl2 treatment
