Abstract
Experiments on culturing isolated meristems of sweet potato (Ipomoea hatatas) were conducted on four local cultivars to overcome a high :incidence of contam:ination and callus:ing of explants. Variables exam:ined :included explant size, cultivar, location of the meristem along the v:ine axis and culture media The aseptic culture of meristems was linproved by: (a) us:ing meristems derived from the tip to the fifth node of v:ines grown :in the greenhouse, not:ing the degree of pubescence of the cultivar to determ:ine the size of meristem to culture; and (b) the use of Murashige and Skoog's nutrient fonnulation + 1 ppm GA3 + 0.1 ppm Ki (WPM medium) or MS + too ml/l coconut water (MSC medium) as culture medium. Growth of isolated meristems was faster on WPM medium but MSC medium can be an alternative when purified growth regulators are not available.
