Abstract
Tissue cultures of maize (Zea mays), wheat (Triticum vulgare), jowar (sorghum vulgare) and rice (Oryza sativa) were established and subcultured for periods ranging from 1 to 3 years. The cultures grew as mixtures of callus and roots on semi-solid media. In liquid shake cultures and growth rate was very high and the cultures grew mainly as roots. The issues grew equally well in glucose, sucrose or starch. In addition to inorganic salts, trace elements and vitamins, the only requirements for growth and viability on repeated subculture were protein hydrolysate and an auxin (naphthalene acetic acid).
