Abstract
The behaviour of pollen from Z. mays, S. bicolor and P. americanum on the stigma of Z. mays was studied. At normal growing temperatures (27-29°C), atmospheric humidity was the main factor controlling germination and pollen-tube penetration; best results were obtained with RH more than 70%. Stigma trichomes provided the best site for pollen attachment, hydration, germination and penetration. In controlled, single-pollen grain pollination, the shortest time between attachment to a stigma trichome and germination was 9 min, 1 min 25 s and 1 min 42 s for pollen from Z. mays, S. bicolor and P. americanum, respectively, in RH 70%. The conformation of the basal cell complex of the trichome determined the direction in which tubes grew into the stigma axis; it was more effective with Z. mays tubes than those of the other species. In the stigma axis, Z. mays pollen tubes reached the transmitting tract by growing through the intercellular spaces of the cortex. S. bicolor tubes failed to reach the tract, but grew fast and were longer in the Z. mays stigma that they are in the S. bicolor stigma. P. americanum tubes penetrated the stigma with difficulty, grew slowly and did not seek the tract. Penetration by tubes of all these species promoted secretion of pectic polysaccharide and protein by stigma axis cells. [See also PBA 54, 2639.]
