Abstract

Allele-specific PCR (AS-PCR) with 3 primers was used to identify true F1 hybrids resulting from normal oleate groundnut cultivars pollinated with high oleate groundnut genotypes with a 1-bp insertion (A:T) at position 442 after the start codon in FAD2B. Its reliability was confirmed by direct sequencing of FAD2B PCR products. The AS-PCR protocol requires minimal resources, and thus provides an economical alternative to qPCR and CAPS methods.
