Abstract

Mature seeds of three varieties were treated with 14 or 26 kR gamma rays or an 0.4% solution of ethylmethanesulphonate (EMS) for 24 h. In a fourth variety, EMS treatments of 0.4 and 0.6% (24 h) of developing embryos (DE) were compared with 0.4% (24 h) treatments of mature seeds (MS). In the first experiment gamma rays gave a higher mutation rate than EMS. In the 26 kR treatment, M1 germination was lower but the M2 mutation frequency was higher. In erect Congo and Dixie Anak, mutations for reduced plant size were most common, followed by chlorophyll mutations, while growth-habit mutations were less frequent. In trailing TBR(V4) all three MS treatments produced a high frequency of erect mutations, the highest rates being obtained with gamma rays. In the second experiment DE treatments with EMS in variety S9 gave more than twice the mutation rate of the MS treatments. In DE treatments, chlorophyll and reduced plant-size mutations were nearly equal in frequency, constituting about 85% of all mutations. In the MS treatments, chlorophyll mutations were more frequent than reduced plant-size mutations, together constituting 94% of the total mutations. Mutations affecting leaflets, which were five times more common in the DE treatments, accounted for most of the differences. There were differences between the DE and MS treatments also in the frequencies of albino, virescent and xantha mutants. Chimera size in the DE treatments was markedly larger
