Abstract
A virus causing wide chlorotic ringspot associated with chlorotic line pattern and mottling was discovered on an Erictoides groundnut hybrid in a greenhouse in Stillwater, Oklahoma, USA. The virus was isolated and characterized and found to differ in symptomatology, host range and serological properties from all previously described viruses infecting groundnuts. The virus was transmitted by mechanical inoculation and grafting to 14 groundnut cultivars, causing symptoms identical to those originally observed. The virus also systemically infected pea cv. Little Marvel and Lupinus albus cv. Tiftwhite. The virus did not infect soyabeans. Phaseolus vulgaris cv. Topcrop and Chenopodium amaranticolor were useful hosts for local lesion assays and for diagnosis. The virus, designated PCRV, had a dilution end point between 10-5 and 10-6, thermal inactivation point between 55 and 60°C, and longevity in vitro up to 6 d. Virus particles observed by EM were flexuous and filamentous, with length 750-850 nm. In serological tests, PCRV was related to a peanut mottle potyvirus isolate from Oklahoma but not to 7 other potyviruses and poty-like viruses tested
